3-O-Sulfation of Heparan Sulfate Enhances Tau Interaction and Cellular Uptake

Prion-like transcellular spreading of tau in Alzheimer's Disease (AD) is mediated by tau binding to cell surface heparan sulfate (HS). However, the structural determinants for tau–HS interaction are not well understood. Microarray and SPR assays of structurally defined HS oligosaccharides show that a rare 3-O-sulfation (3-O-S) of HS significantly enhances tau binding. In Hs3st1^−/− (HS 3-O-sulfotransferase-1 knockout) cells, reduced 3-O-S levels of HS diminished both cell surface binding and internalization of tau. In a cell culture, the addition of a 3-O-S HS 12-mer reduced both tau cell surface binding and cellular uptake. NMR titrations mapped 3-O-S binding sites to the microtubule binding repeat 2 (R2) and proline-rich region 2 (PRR2) of tau. Tau is only the seventh protein currently known to recognize HS 3-O-sulfation. Our work demonstrates that this rare 3-O-sulfation enhances tau–HS binding and likely the transcellular spread of tau, providing a novel target for disease-modifying treatment of AD and other tauopathies.     

The Green polynomials via vertex operators

An iterative formula for the Green polynomial is given using the vertex operator realization of the Hall-Littlewood function. Based on this, (1) a general combinatorial formula of the Green polynomial is given; (2) several compact formulas are given for Green's polynomials associated with upper partitions of length ≤3 and the diagonal lengths ≤3; (3) a Murnaghan-Nakayama type formula for the Green polynomial is obtained; and (4) an iterative formula is derived for the bitrace of the finite general linear group G and the Iwahori-Hecke algebra of type A on the permutation module of G by its Borel subgroup.     

19F-detected dual-optimized inverted 1JCC 1,n-ADEQUATE

Modification of the recently reported ¹⁹F-detected 1,1-ADEQUATE experiment that incorporates dual-optimization to selectively invert a wide range of ¹J_CC correlations in a 1,n-ADEQUATE experiment is reported. Parameters for the dual-optimization segment of the pulse sequence were modified to accommodate the increased size of ¹J_CC homonuclear coupling constants of poly- and perfluorinated molecules relative to protonated molecules to allow broadband inversion of the ¹J_CC correlations. The observation and utility of isotope shifts are reported for the first time for 1,1- and 1,n-ADEQUATE correlations.     

An Infinite Antichain of Planar Tanglegrams

Contrary to the expectation arising from the tanglegram Kuratowski theorem of Czabarka et al. (SIAM J. Discrete Math. 31(3), 1732–1750, 2017), we construct an infinite antichain of planar tanglegrams with respect to the induced subtanglegram partial order. R.E. Tarjan, R. Laver, D.A. Spielman and M. Bóna, and possibly others, showed that the partially ordered set of finite permutations ordered by deletion of entries contains an infinite antichain, i.e., there exists an infinite collection of permutations, such that none of them contains another as a pattern. Our construction adds a twist to the construction of Spielman and Bóna (Electr. J. Comb. 7, N2, 2000).

     

A category of kernels for equivariant factorizations,II: Further implications

We leverage the results of the prequel [8], in combination with a theorem of D. Orlov to create a categorical covering picture for factorizations. As applications, we provide a conjectural geometric framework to further understand M. Kontsevich's Homological Mirror Symmetry conjecture and obtain new cases of a conjecture of Orlov concerning the Rouquier dimension of the bounded derived category of coherent sheaves on a smooth variety.
